Bifidobacterium Medium Recipe

1. Dissolve the following in 1 L of distilled water to make Salt Solution:
• NaCl 2.00 g
• NaHCO₃ 10.00 g
• KH₂PO₄ 1.00 g
• K₂HPO₄ 1.00 g
• MgSO₄ x 7 H₂O 0.50 g
• CaCl₂ x 2 H₂O 0.25 g
2. Dissolve the following in 950 ml of distilled water:
• Resazurin (25 mg/100 ml) 4 ml
• Salt solution (see above) 40 ml
• NaCl 5.0 g
• Tween 80 1.0 ml
• MnSO₄ x H₂O 0.05 g
• MgSO₄ x 7 H₂O 0.2 g
• K₂HPO₄ 2.0 g
• Glucose 10.0 g
• Bacto Soytone 5.0 g
• Yeast extract 5.0 g
• Meat extract 5.0 g
• Casein peptone, tryptic digest 10.0 g
3. Boil medium and cool under CO2.
4. Add Cysteine-HCl x H₂O 0.5 g to medium
5. Adjust pH to 6.8 using 8 N NaOH.
6. Distribute under N₂ and autoclave to sterilize.
