Desulfovibrio Medium Recipe

  1. Dissolve the following in 800 ml of distilled water to make Solution A:
    • NH4Cl 1.0 g
    • K2HPO4 0.5 g
    • Resazurin 1.0 mg
    • Yeast extract 1.0 g
    • DL-Na-lactate 2.0 g
    • MgSO4 x 7 H2O 2.0 g
    • CaCl2 x 2 H2O 0.1 g
    • Na2SO4 1.0 g
  2. q.s. to 980 ml with distilled water
  3. Dissolve the following in 10 ml of distilled water to make Solution B:
    • FeSO4 x 7 H2O 0.5 g
  4. Dissolve the following in 10ml of distilled water to make Solution C:
    • Ascorbic acid 0.1 g
    • Na-thioglycolate 0.1 g
  5. Bring solution A to the boil for a few minutes, then cool to room temperature while gassing with oxygen-free N2 gas.
  6. Add solution B and C, adjust pH to 7.8 with NaOH, and distribute under N2 in anaerobic tubes.
  7. During distribution continuously swirl the medium to keep the grey precipitate suspended.
  8. Autoclave to sterilize.
