Starch - Mineral Salt (STMS) Agar Recipe

  1. Dissolve the following in 100 ml of distilled water to make trace elements solution:
    • ZnSO4 x 7 H2O 0.1 g
    • MnCl2 x 4 H2O 0.1 g
    • FeSO4 x 7 H2O 0.1 g
  2. Add 20g of of rolled oats to 900 ml distilled water
    • Agar 15.0 g
    • Trace element solution (see above) 1.0 ml
    • CaCO3 2.0 g
    • NaCl 1.0 g
    • MgSO4 x 7 H2O 1.0 g
    • K2HPO4 1.0 g
    • (NH4)2SO4 2.0 g
    • Starch, soluble 10.0 g
  3. q.s. to 1L with distilled water.
  4. Adjust pH to 7.2.
  5. Autoclave to sterilize.
